چکیده
Campylobacter remains the leading cause responsible of human gastroenteritis worldwide. The current study aimed to compare internal DNA extraction protocol with two commercially available kits, using C. jejuni (ATCC® 29428TM) and coli (ATCC®43478TM) validate optimize through artificial contamination confirmation spp. from broiler chickens, turkeys, beef meats samples. processes were carried out following protocols steps manufacturer's instructions PureLinkTM Genomic Mini Kit Wizard® Purification Kit, respectively. agarose gel electrophoresis system was used control quality. After that, forty isolates confirmed, finally, an aforementioned reference strains at three different concentrations performed sterilized minced meats. This is first work comparing genomic for results indicated that provided similar efficiency kits. All confirmed successfully amplified, in which28 12 jejuni, as revealed by biochemical tests. A positive amplification also observed contaminated food matrices, after enrichment, all examined doses. Except some reactions negative 1 CFU/mL coli. explained detection limits both qPCR. Based on our findings, crucial determining quality this amended. Hence, highlighted importance validating simpler, cheaper faster protocol, each laboratory, part future risk assessment, monitoring programs frequency required molecular studies.

Optimization of the genomic DNA extraction in some mosses
The presence of organic compounds and high amount of secondary metabolites (polysaccharides, phenolic component, etc.) in mosses cause difficulties in DNA extraction that are followed by problems in PCR reactions. In lower plants, various methods have been used for DNA extraction including silica gel and different commercial kits. These methods mostly use hazardous (like phenol or liquid nitrog...

DNA extraction from plant tissues often causes most problems. For example, unsuccessful removal secondary metabolites during extraction, such as phenolic compounds in aromatic and medicinal plants, cause to some mistakes in result of molecular experiments by using of the extracted DNA. Achillea wilhelmsii is a medicinal plant belong to Asteraceae family and native to Iran, there is little infor...

Modified Cetyltrimethylammonium bromide (CTAB) protocol for DNA isolation was developed from leaf tissuesof Phyllanthus emblica for obtaining high quality genomic DNA. Fresh leaves of three different maturitywere analyzed for yield and quality of DNA. Acidity was determined in three different maturity of leaves viz.tender, intermediate and mature and their influence on DNA qua...
متن کاملProtocol optimization for genomic DNA extraction and RAPD-PCR in mosquito larvae (Diptera: Culicidae)
PCR based molecular markers are powerful tools for the analysis of genetic diversity for which isolation of good quality genomic DNA is essential. This paper presents a comparison of three DNA extraction methods for 1, 5 and 10 larvae of Aedes aegypti. First method employing DNAzol® involves simple and one step protocol for DNA isolation where maximum purity (A260/A280 1.9) was obtained with si...
متن کاملProtocol for extraction of genomic DNA from swine solid tissues
Molecular diagnostics are performed by using DNA from different body tissues. However, it is necessary to obtain genomic DNA of good quality. Due to the impossibility of collecting blood from slaughtered animals, DNA extraction from solid tissues is necessary. The objective of this study was to describe a protocol of DNA extraction from swine skin, adipose, brain, liver, kidney and muscle tissu...
